First up come the produce, flowers, and souvenir stalls flanking Ruga d. Oréfici, filling Campo San Giacomo Rialto, and (after a dogleg right then left again) Campo Bella Vienna.
If you wend your way through these stalls (and remember to dogleg right) you will eventually find the famous Pescheria, a Renaissance loggia right on the Grand Canal that for centuries has shaded the fish vendors of Venice. (Given that it is seafood, the action is at its height in the morning.)
(I know no one saw the movie, but it is over the roofs of the fish market stalls extending from the loggia that the Johnny Depp character makes his escape in The Tourist—a movie seemingly made just so Depp and Angelina Jolie could hang out in Venice for a few weeks.)
Mercato del Rialto (Rialto Market)
Ruga d. Oréfici/Campo San Giacomo Rialto/Campo Bella Vienna/Campo della Pescheria (S. Polo).
Vaporetto: Rialto

Daily (Produce: Mon–Sat 7am–8pm; Fish: Tues–Sat 7am–2pm)
Planning your day: It's a market. Spend as long as you like. It woudl probably take about 15–30 minutes to stroll all the way through, pausing to take photographs and maybe haggle a bit. Venice itineraries
Being a food market neighborhod, there are a number of good restaurants (I recommend Viini Da Pinto, overlooking the Pescheria) and, particualrly, cicchetti bars in this part of San Polo (the best: Cantina Do Mori).
